The A64 was blocked between the Grimston and Hopgrove roundabouts following an accident between three vehicles on the eastbound carriageway.
Emergency services are currently in attendance and the Yorkshire Air Ambulance was called.
One casualty has been taken to hospital. One lane is open eastbound, and the westbound carriageway is clear.
Police are advising drivers that there will be delays for some time.
